My current research focuses on understanding how different DNA-binding proteins gain access to their specific target sequences located in the nucleosome, which not only acts as a fundamental packaging unit of genome organization but also contributes to gene expression. Using first-passage probability approaches, we aim to develop stochastic models to investigate how different nucleosome dynamics such as DNA breathing, sliding, or repositioning affect the target search dynamics of DNA-binding proteins using theoretical and computational methods. Such models not only have the potential to provide a clear physical-chemical picture of complex biological processes but can also be utilized in designing new experiments.
